Hope Rugo, MD, UCSF Helen Diller Family Comprehensive Cancer Center, San Francisco, CA, discusses the potential of novel agents in combination therapy for breast cancer. The I-SPY 2.2 trial (NCT01042379) demonstrated high pathologic complete response (pCR) rates in patients with high-risk disease who received datopotamab deruxtecan, a TROP2-directed antibody-drug conjugate (ADC) and durvalumab. For patients who did not meet the pCR criteria, they received the KEYNOTE-522 trial (NCT03036488) regimen of pembrolizumab with neoadjuvant chemotherapy, and subsequently have promising pCR rates. This interview took place at the 2025 St. Gallen International Breast Cancer Consensus Meeting in Vienna, Austria.
