In this roundtable discussion, J. Randolph Hecht, MD, UCLA Santa Monica Medical Center, Santa Monica, CA, is joined by Jennifer Specht, MD, Fred Hutch Cancer Center, Seattle, WA, Winfried Alsdorf, MD, University Medical Center Hamburg-Eppendorf, Hamburg, Germany, and Susanne Baumeister, MD, Dana-Farber Cancer Institute, Boston, MA, to discuss advancements in CAR-T therapies targeting cell surface proteins in solid tumors. Dr Hecht presents updates on logic-gated CAR-T therapies for pancreatic cancer, while Dr Specht discusses CAR-T applications in breast and ovarian cancers. Dr Alsdorf introduces CLDN6-targeted CAR-T therapy for relapsed germ cell tumors, and Dr Baumeister explores ALK CAR T cells for relapsed/refractory (R/R) neuroblastoma. The panel addresses challenges in antigen selection, tumor heterogeneity, and strategies to enhance CAR-T cell persistence and efficacy in solid tumors. This discussion took place at the 7th International Workshop on CAR-T and bispecifics (iwCAR-T) 2025 held in Miami, FL.
