Dean Fennell, FRCP, PhD, University of Leicester, Leicester, UK, comments on the Phase I MITOPE trial (NCT05278975), evaluating the safety and maximum tolerated dose (MTD) of intra-pleural RSO-021 in patients with mesothelioma or other pleural cancers. RSO-021, a cyclic oligopeptide targeting mitochondrial PRX3, demonstrated signs of anti-tumor activity and manageable toxicity at 90 mg/week, with common side effects including fatigue, pyrexia, and elevated creatinine. The trial supports further Phase II studies of RSO-021 as a monotherapy and in combination with other therapies.
