Whilst CAR T-cells have been relatively successful in hematological malignancies, barriers to their adoption in solid tumors have been partially due to suitable target selection. Targeting common receptors such as HER2 may result in serious adverse events. Benjamin Schlechter, MD, Dana-Farber Cancer Institute, Boston, MA, describes the rationale behind the development of TAC T-cell therapies, which can reduce off-tumor toxicity compared to earlier generations of CAR T-cells.
