Alberto Montero, MD, Seidman Cancer Center at University Hospitals, Cleveland, OH, discusses the initial findings of a Phase I/II study (NCT05768139) of STX-478, a mutant-selective PI3Kα inhibitor, in patients with advanced PI3Kα mutant solid tumors. STX-478, designed to minimize wild-type (WT) toxicities, demonstrated robust efficacy and tolerability in heavily pre-treated patients, including those with prior PI3K inhibitor intolerance. The novel PI3Kα inhibitor has additionally shown promise in terms of fewer toxicities and potential combination strategies. This interview took place at the European Society for Medical Oncology (ESMO) 2024 Congress in Barcelona, Spain.
