Bart Neyns, MD, PhD, Universitair Ziekenhuis Brussel, Brussels, Belgium, discusses the rationale for using conventional dendritic cells in cellular therapy for recurrent glioblastoma. Preclinical models indicate that these dendritic cells, which the tumor microenvironment of glioblastoma lack, are crucial for effective anti-cancer immune responses, including immune checkpoint inhibitors and adoptive T-cell therapies. Following promising results in checkpoint-refractory solid tumors, a Phase I trial (NCT03233152) in patients with glioblastoma demonstrated the feasibility and safety of combining dendritic cells with nivolumab and ipilimumab.
