SITC 2024 | Intratumoral Fc-optimized CD40 antibody induces tumor rejection & systemic immunity in met cancer

Juan Osorio, MD, Memorial Sloan Kettering Cancer Center, The Rockefeller University, New York, NY, shares the preliminary findings from the in-human Phase I study (NCT04059588) investigating the clinical activity and tolerability of 2141-V11, an anti-CD40 antibody, through dose-escalation. 12 patients with identifiable metastatic lesions for intratumoral injections were given 4 different dose levels once every 3 weeks. 2141-V11 was found to be safe and efficacious, wherein 2 patients showed a complete response, and 6 patients had significant reductions in tumor burden. Post-treatment biopsies additionally revealed immune infiltrations in local and distal lesions, which has paved the way for further clinical studies in various tumor types exploring the promising clinical efficacy of 2141-V11.
