ESMO Breast 2025 | DESTINY-Breast07: updated results of first line T-DXd in HER2+ breast cancer

Fabrice André, MD, PhD, Gustave Roussy, Villejuif, France, presents updated results from the DESTINY-Breast07 trial (NCT04538742), a Phase Ib/II study assessing the safety and efficacy of trastuzumab deruxtecan (T-DXd), with or without pertuzumab, in the first-line setting for patients with HER2-positive metastatic breast cancer. A high overall response rate and prolonged progression-free survival (PFS) across the cohort was reported. Exploratory molecular subgroup analyses did not reveal any clear predictors of differential benefit. These findings support further investigation in the Phase III DESTINY-Breast09 trial (NCT04784715), with results anticipated soon. This interview took place at the 2025 European Society for Medical Oncology (ESMO) Breast Cancer Annual Congress in Munich, Germany.
