ESMO 2025 | BURAN: buparlisib and paclitaxel in pre-treated HNSCC

Denis Soulieres, MD, Centre Hospitalier de l’Université de Montréal, Montreal, Canada, discusses the Phase III BURAN study (NCT04338399) of buparlisib plus paclitaxel versus paclitaxel alone in patients with recurrent or metastatic head and neck squamous cell carcinoma (HNSCC) previously treated with PD-1/PD-L1 inhibitors. The addition of buparlisib did not improve overall survival or progression-free survival compared with paclitaxel alone, though a modest increase in response rate was observed. Safety findings were consistent with prior studies, with no new safety signals identified. This interview took place at the European Society for Medical Oncology (ESMO) 2025 Congress in Berlin, Germany.
