ELCC 2026 | Calderasib, a novel KRAS G12C-off inhibitor in NSCLC: insights from the KANDLELIT-001 trial
Adrian Sacher, MD, MMSc, FRCPC, Princess Margaret Cancer Centre, Toronto, Canada, presents data from the KANDLELIT-001 trial (NCT05067283) evaluating a novel KRAS G12C-off inhibitor calderasib alone and in combination with pembrolizumab or chemotherapy, in previously treated and in treatment-naïve patients with metastatic G12C-mutated non-small cell lung cancer (NSCLC). In previously treated patients, calderasib demonstrated clinically significant, fairly durable response rates in combination and alone, however the response rates for calderasib alone were modest. In PD-L1 high patients, a response rate of 87% was achieved, with the median progression-free survival not yet reached for this cohort. The combinations were tolerable, with modest rates of autoimmune hepatitis. This interview took place at the 2026 European Lung Cancer Congress in Copenhagen, Denmark.
