ESMO GI 2026 | HERIZON-GEA-01: response depth and PFS2 in HER2+ gastroesophageal cancer

Filippo Pietrantonio, MD, Fondazione IRCCS Istituto Nazionale dei Tumori, Milan, Italy, discusses a post hoc analysis of the HERIZON-GEA-01 trial (NCT05152147), examining response characteristics and progression-free survival after subsequent therapy (PFS2) among treatment responders receiving zanidatamab plus chemotherapy, with or without tislelizumab, versus trastuzumab plus chemotherapy in HER2-positive advanced gastroesophageal adenocarcinoma. No baseline characteristics predicted longer duration of response. Adding tislelizumab appeared to contribute to treatment effect, with greater tumor shrinkage and longer progression-free survival, while prolonged PFS2 across experimental arms suggested continued benefit from subsequent therapies. Safety was consistent with the overall population. This interview took place at the European Society for Medical Oncology (ESMO) Gastrointestinal Cancer 2026 Congress in Munich, Germany.
