SITC 2024 | Phase Ib trial results on TLS density pre- and post-oncolytic immunotherapy with ICIs for MIBC

Lee Moffitt Cancer Center, Tampa, FL, shares results from a Phase Ib study (NCT04610671) investigating the density of tertiary lymphoid structures (TLS) before and after combination therapy with intravesical cretostimogene grenadenorepvec and systemic nivolumab in patients with muscle-invasive bladder cancer (MIBC). High-density tertiary lymphoid structures (TLS) at baseline did not predict complete response, however, TLS formation was seen in patients who achieved a complete response post-treatment. High levels of CD138-positive plasma cells indicated the increasing density and maturity of TLS within the tumor and surrounding stroma. Isotype switching from IgM antibodies at baseline to IgA/IgG antibodies post-treatment was also observed in tumor cells of patients with a complete pathologic response.
