ASCO 2025 | Phase I trial of a therapeutic vaccine with checkpoint inhibitors for FLC

Margaret Gatti-Mays, MD MPH, The Ohio State University, Columbus, OH, shares findings from a Phase I trial (NCT04248569) of a therapeutic vaccine targeting the DNAJ-PKAc chimera with nivolumab and ipilimumab for advanced fibrolamellar hepatocellular carcinoma (FLC). The study demonstrated acceptable safety and immunogenicity, and clinical benefit was observed in a substantial proportion of evaluable patients, with evidence suggesting a link between T cell reactivity and disease control. T cell exhaustion emerged as a mechanism of resistance in some cases. This interview took place during the 2025 American Society of Clinical Oncology (ASCO) Meeting in Chicago, IL.
