ESMO 2024 | Neo-CheckRay: boosting pCR rates in cold tumors

Alex De Caluwe, MD, Jules Bordet Institute, Brussels, Belgium, discusses the Phase II Neo-CheckRay trial (NCT03875573), which combines radiotherapy and immunotherapy in patients with early-stage, high risk ER+/HER2- breast cancer. The trial showed impressive results, where addition of durvalumab increased residual cancer burden (RCB) 0-1 rates and pathological complete response (pCR) rate. Notably, a increase in pCR was observed in cold, PD-L1-negative tumors, suggesting that radiotherapy may enhance immunotherapy effectiveness in these harder-to-treat cancers. The trial points toward future strategies of reducing radiation doses while maintaining high efficacy. This interview took place at the European Society for Medical Oncology (ESMO) 2024 Congress in Barcelona, Spain.
