Rupert Bartsch, MD, Medical University of Vienna, Vienna, Austria, gives an overview of the the EMBER-3 (NCT04975308) and PADA-1 (NCT03079011) trials in hormone receptor-positive, HER2-negative metastatic breast cancer. EMBER-3 evaluated imlunestrant with or without abemaciclib in patients progressing on prior CDK4/6 inhibitors, and consistent progression-free survival benefit across subgroups was reported. PADA-1 assessed blood ESR1 mutation emergence during aromatase inhibitor and palbociclib therapy, identifying clinical factors associated with mutation kinetics. This interview took place at the 2025 European Society for Medical Oncology (ESMO) Breast Cancer Annual Congress in Munich, Germany.
