Rupert Bartsch, MD, Medical University of Vienna, Vienna, Austria, discusses the post-hoc subanalysis of the Phase II TUXEDO-3 trial (NCT05865990) of patritumab deruxtecan (HER3-DXd) in patients with HER2-positive metastatic breast cancer and active brain metastases or leptomeningeal disease previously treated with trastuzumab deruxtecan (T-DXd). In this heavily pretreated population, HER3-DXd demonstrated encouraging intracranial and bicompartmental progression-free survival (PFS) exceeding 6 months, supporting the feasibility of sequential antibody-drug conjugate (ADC) strategies in central nervous system (CNS) disease.
