Erika Hamilton, MD, Sarah Cannon Research Institute, Nashville, TN, discusses the Phase III HER2CLIMB-05 study (NCT05132582), which evaluated the addition of tucatinib to trastuzumab and pertuzumab as first-line (1L) maintenance therapy for HER2+ metastatic breast cancer. Tucatinib significantly improved progression-free survival versus placebo, with consistent benefit across all subgroups, including those with brain metastases. Early overall survival data trend in favor of tucatinib, and safety was manageable, with diarrhea and elevated liver enzymes as the most common adverse events.
