ASCO 2026 | Phase III MACE trial: metronomic chemotherapy after surgery in oral cavity squamous cell carcinoma

Nandini Menon, MBBS, MD, DNB, Tata Memorial Centre, Mumbai, India, discusses the results of a Phase III randomized controlled trial investigating metronomic adjunct chemotherapy (MACE) in locally advanced oral cavity squamous cell carcinoma post-surgery and adjuvant treatment, where patients received low-dose methotrexate and celecoxib. Dr Menon highlights that although the study did not meet statistical significance due to being underpowered, there was a numerical increase in overall survival with metronomic chemotherapy, particularly in high-risk subgroups, suggesting a potential signal that warrants further investigation. This interview took place during the 2026 American Society of Clinical Oncology (ASCO) Meeting in Chicago, IL.
