ASCO 2026 | EV-202 cohort 9: enfortumab vedotin + pembrolizumab as first-line treatment in PDL-1+ R/M HNSCC

Ari Rosenberg, MD, University of Chicago, Chicago, IL, discusses the results of the Phase II EV-202 study (NCT04225117), which investigated the combination of enfortumab vedotin and pembrolizumab for frontline PD-L1-positive recurrent or metastatic head and neck squamous cell carcinoma (R/M HNSCC). He notes that the objective response rate (ORR) was 39%, with responses enriched for HPV-positive patients. Dr Rosenberg also highlights the potential of antibody drug conjugates (ADCs) targeting Nectin-4, particularly in patients with oropharynx primary or HPV-positive tumors. This interview took place during the 2026 American Society of Clinical Oncology (ASCO) Meeting in Chicago, IL.
