Giulia Martini, MD, PhD, Università degli Studi della Campania Luigi Vanvitelli, Naples, Italy, xplores results from the CAPRI-2 GOIM trial (NCT05312398), evaluating plasma circulating tumor DNA comprehensive genomic profiling to guide sequential cetuximab-based therapy in RAS/BRAF wild-type, microsatellite stable metastatic colorectal cancer. Patients without resistance-associated variants (“negative hyperselected”) had significantly better response rates and progression-free survival with continued cetuximab across first- and second-line treatment compared with those harboring resistance variants. Findings support using liquid biopsy profiling to identify patients likely to benefit from ongoing anti-epidermal growth factor receptor therapy.
