WCLC 2026 | Class 3 BRAF alterations predict poor immunotherapy outcomes in NSCLC

Alessandro Di Federico, MD, Memorial Sloan Kettering Cancer Center, New York, NY, discusses a retrospective study on class 2 and class 3 BRAF alterations in non-small cell lung cancer (NSCLC), which found that class 3 alterations were associated with a higher tumor mutational burden and poorer outcomes with immunotherapy. Mutations in genes such as STK11, CDKN1, and SMARCA4 were more frequent in tumors with class 3 alterations and were consistently associated with worse overall survival, suggesting that these tumors may have limited responsiveness to immunotherapy. This interview took place at the IASLC 2026 World Conference on Lung Cancer congress in Seoul, Republic of Korea.
