ELCC 2026 | Current and emerging HER2-targeted therapies in NSCLC
Nicolas Girard, MD, PhD, Institut Curie, Paris, France, explores current and emerging therapies for HER2-mutated non-small cell lung cancer (NSCLC). The antibody-drug conjugate trastuzumab deruxtecan (T-DXd) is being investigated in an ongoing DESTINY-Lung04 study (NCT05048797) as a first-line treatment, and previous data for T-DXd in later line settings show a response rate of around 50% and a progression-free survival of around 12 months. Dr Girard also discusses promising data from the HER2-directed tyrosine kinase inhibitors zongertinib and sevabertinib, showing higher response rates of 70% and prolonged duration of responses. However, the safety profile has promoted concern, with high rates of diarrhea, although most of these events have been low grade. This interview took place at the 2026 European Lung Cancer Congress in Copenhagen, Denmark.
