Alessandro Di Federico, MD, Memorial Sloan Kettering Cancer Center, New York, NY, discusses a retrospective study on class 2 and class 3 BRAF alterations in non-small cell lung cancer (NSCLC), which found that class 3 alterations were associated with a higher tumor mutational burden and poorer outcomes with immunotherapy. Mutations in genes such as STK11, CDKN1, and SMARCA4 were more frequent in tumors with class 3 alterations and were consistently associated with worse overall survival, suggesting that these tumors may have limited responsiveness to immunotherapy.
