ESMO 2025 | HLA-restricted Immunotherapies and resistance mechanisms in cancer

Michael Gormally, MD, PhD, Memorial Sloan Kettering Cancer Center, New York, NY, discusses emerging progress in HLA-restricted immunotherapies, highlighting recent regulatory approvals, including the bispecific T-cell engager tebentafusp for uveal melanoma and the adoptive cell therapy afamitresgene autoleucel (afami-cel) for synovial sarcoma. Dr Gormally explores mechanisms of resistance, focusing on HLA loss of heterozygosity (LOH) and its relationship to neoantigen burden in tumors with KRAS and TP53 mutations. This interview took place at the European Society for Medical Oncology (ESMO) 2025 Congress in Berlin, Germany.
