Pamrevlumab, a humanized monoclonal antibody targeting connective tissue growth factor (CTGF), plays a crucial role in modulating tumor-stroma interactions in pancreatic cancer. Vincent Picozzi, MD, Virginia Mason Medical Center, Seattle, WA, describes earlier preclinical studies which demonstrate pamrevlumab’s potential to inhibit cancer cell growth in both cell lines and genetically engineered mouse models. Further research showed promise when combined with gemcitabine and nab-paclitaxel, suggesting an increased likelihood of surgical resection in patients with locally advanced pancreatic cancer. These findings provided foundations for an arm of the ongoing Precision Promise trial (NCT04229004) evaluating multiple experimental treatments, including pamrevlumab, in patients with metastatic pancreatic cancer.
