ASCO 2025 | Biomarkers predicting treatment benefit in BRAF-mutated NSCLC

Alessandro Di Federico, MD, Dana-Farber Cancer Institute, Boston, MA, identifies key biomarkers predicting treatment benefit in BRAF-mutated lung cancer patients, as seen in the FRONT-BRAF study. Tobacco use history and PD-L1 expression (≥1%) drive survival advantages with immunotherapy, confirmed through statistical adjustments including propensity score matching. TP53 co-mutation emerges as another crucial biomarker, where patients with both BRAF and TP53 mutations show significantly longer overall survival with immunotherapy, while TP53 wild-type patients benefit more from targeted therapy with longer progression-free survival. This interview took place during the 2025 American Society of Clinical Oncology (ASCO) Meeting in Chicago, IL.
