ASCO 2025 | Early Phase I findings for B7-H4-directed ADC Emi-Le in solid tumors

Erika Hamilton, MD, Sarah Cannon Research Institute, Nashville, TN, discusses initial dose escalation results from the Phase I trial (NCT05377996) of emiltatug ledadotin (Emi-Le), a B7-H4-directed dolasynthen antibody-drug conjugate (ADC), in patients with advanced solid tumors. Emi-Le was generally well tolerated, with a manageable safety profile and no unexpected toxicities. Antitumor activity was observed across dose levels, particularly in patients with high B7-H4 expression, including those previously treated with topoisomerase-1 inhibitors. These findings support continued development in B7-H4-positive triple-negative breast cancer (TNBC) and related tumor types. This interview took place during the 2025 American Society of Clinical Oncology (ASCO) Meeting in Chicago, IL.
