Filippo Pietrantonio, MD, Fondazione IRCCS Istituto Nazionale dei Tumori, Milan, Italy, discusses a post hoc analysis of the HERIZON-GEA-01 trial (NCT05152147), examining response characteristics and progression-free survival after subsequent therapy (PFS2) among treatment responders receiving zanidatamab plus chemotherapy, with or without tislelizumab, versus trastuzumab plus chemotherapy in HER2-positive advanced gastroesophageal adenocarcinoma. No baseline characteristics predicted longer duration of response. Adding tislelizumab appeared to contribute to treatment effect, with greater tumor shrinkage and longer progression-free survival, while prolonged PFS2 across experimental arms suggested continued benefit from subsequent therapies.
