ESMO 2026 | KEYNOTE-975: pembrolizumab + chemoradiotherapy did not improve EFS or OS in GEJ cancer

Filippo Pietrantonio, MD, Fondazione IRCCS Istituto Nazionale dei Tumori, Milan, Italy, discusses the KEYNOTE-975 trial (NCT04210115) investigating pembrolizumab plus concurrent definitive chemoradiotherapy in esophageal and gastroesophageal junction (GEJ) cancer. The trial failed to meet its primary endpoint of improving event-free (EFS) and overall survival (OS). Dr Pietrantonio highlights that the results were not due to increased toxicity, but possibly due to the timing of the intervention, and that other trials are now exploring the role of consolidation immunotherapy after definitive chemoradiation. This interview took place at the European Society for Medical Oncology (ESMO) 2026 Congress in Madrid, Spain.
