ASCO 2025 | DANTE: evaluating 1 vs 2 years of anti-PD-1 therapy in metastatic melanoma

Omid Hamid, MD, The Angeles Clinic and Research Institute, Los Angeles, CA, discusses the Phase III DANTE trial (ISRCTN15837212), which compared 1 year versus at least 2 years of first-line anti-PD-1-based immunotherapy in metastatic melanoma. Although a shorter duration appeared non-inferior for progression-free survival, early trial closure limited statistical power. Findings support continued use of 2-year therapy as standard while ongoing analyses further explore quality of life, toxicity, and cost-effectiveness outcomes. The PET‐Stop trial (NCT04462406) will subsequently assess biomarkers seen on PET/CT imaging to stop therapy in this setting. This interview took place during the 2025 American Society of Clinical Oncology (ASCO) Meeting in Chicago, IL.
