AACR 2025 | ctDNA-guided PD-1 blockade in resected early-stage MMRd tumors

Yelena Janjigian, MD, Memorial Sloan Kettering Cancer Center, New York, NY, discusses a prospective study evaluating pembrolizumab in patients with resected mismatch repair deficient solid tumors and circulating tumor DNA (ctDNA)-detected minimal residual disease (MRD). Patients with positive ctDNA received PD-1 blockade, while ctDNA-negative patients were observed. High rates of ctDNA clearance and promising recurrence-free and overall survival outcomes were observed among treated patients. Findings suggest ctDNA-guided immunotherapy may offer clinical benefit across multiple tumor types following surgery. This interview took place at American Association for Cancer Research (AACR) Annual Meeting 2025 in Chicago, IL.
