Margaret Gatti-Mays, MD MPH, The Ohio State University, Columbus, OH, discusses a Phase Ib/II trial (NCT06026657) of naxitamab, an anti-GD2 monoclonal antibody, gemcitabine, with TGFβ-imprinted, IL-21 expanded NK cells (TGFβi NKs) in patient with HER2-negative breast cancer. By targeting immune suppression in the tumor microenvironment, this combination leverages NK cell activity and antibody-dependent cytotoxicity. Outcomes include response rates, progression-free survival, and immune biomarkers, with early preclinical data showing promising efficacy.
