ASCO 2026 | ctDNA clearance as an early efficacy biomarker in KRAS G12C-mutated mCRC

Filippo Pietrantonio, MD, Fondazione IRCCS Istituto Nazionale dei Tumori, Milan, Italy, provides an overview of early circulating tumor DNA (ctDNA) clearance as a non-invasive biomarker of treatment response in the Phase III CodeBreaK 300 trial (NCT05198934) of sotorasib plus panitumumab in KRAS G12C-mutant metastatic colorectal cancer (mCRC). Early ctDNA clearance was more frequent with sotorasib-based therapy versus investigator’s choice and was prognostic for improved outcomes, supporting its utility as an early response biomarker independent of treatment arm. This interview took place during the 2026 American Society of Clinical Oncology (ASCO) Meeting in Chicago, IL.
