AACR 2025 | Exarafenib shows promise in treating BRAF-mutant NSCLC

Tadashi Manabe, MD, PhD, University of California, San Francisco, CA, discusses the mechanism and preclinical efficacy of exarafenib, a novel pan-RAF inhibitor targeting both monomeric and dimeric RAF forms in BRAF-mutated non-small cell lung cancer (NSCLC). This includes Class II and III mutations, which currently lack approved targeted therapies and are linked to poorer outcomes. Exarafenib demonstrated potent MAPK pathway inhibition, strong anti-tumor activity, and favorable pharmacologic properties. This interview took place at American Association for Cancer Research (AACR) Annual Meeting 2025 in Chicago, IL.
