ASCO 2026 | Resistance mechanisms following first-line lorlatinib: insights from the CROWN trial

Tony Mok, MD, Chinese University of Hong Kong, Hong Kong, China, discusses that in the Phase III CROWN study (NCT03052608), following first-line lorlatinib, no L-dependent resistance mechanisms were observed, however they identified RAS pathway mutations and tumor suppressor genes. Prof. Mok highlights that these findings do not provide sufficient evidence to inform treatment choices for patients. This interview took place during the 2026 American Society of Clinical Oncology (ASCO) Meeting in Chicago, IL.
