Timothy Yap, MBBS, PhD, FRCP, The University of Texas MD Anderson Cancer Center, Houston, TX, comments on findings from the first-in-human trial (NCT06004245) of RO7589831, a WRN inhibitor, in patients with advanced microsatellite instability (MSI) and/or mismatch repair-deficient (dMMR) solid tumors. RO7589831 demonstrated a favorable safety profile and early signals of efficacy, including partial responses and metabolic activity in post-immunotherapy settings. Durable disease control and ctDNA responses support its potential as a targeted approach for patients with limited options following immune checkpoint inhibitor failure. Dose optimization is ongoing.
